Intracytoplasmic Sperm Injection (ICSI)
A cutting-edge micromanipulation technique where a single, hand-selected sperm is directly injected into the cytoplasm of a mature egg.
Treatment Highlights & Advantages
What is Intracytoplasmic Sperm Injection (ICSI)?
Intracytoplasmic Sperm Injection (ICSI) is an advanced micromanipulation breakthrough that has transformed the management of male factor infertility. While traditional IVF places thousands of sperm around an egg in a petri dish hoping one penetrates naturally, ICSI involves our senior clinical embryologists meticulously selecting a single, morphologically flawless sperm and injecting it directly into the cytoplasm of a mature egg.
Under the leadership of our senior embryology director Dr. Ssuniti, Krisna IVF utilizes motorized micro-injectors, anti-vibration tables, and high-definition inverted optics. This delicate procedure maximizes fertilization probability even when sperm numbers are critically low or when sperm are retrieved surgically via TESA (Testicular Sperm Aspiration) or PESA (Percutaneous Epididymal Sperm Aspiration).
The ICSI Step-by-Step Procedure
Ovarian Stimulation & Retrieval
The female partner undergoes controlled stimulation followed by ultrasound-guided egg retrieval. In the embryology lab, the surrounding cumulus cells are enzymatically stripped to verify metaphase II (MII) maturity.
High-Resolution Sperm Selection
Under high magnification (400x to 1000x), individual sperm are evaluated for head symmetry, intact acrosome, and progressive motility. The tail is gently immobilized using a glass micropipette to initiate activation.
Micro-Injection of Oocyte
The mature egg is held firmly by a gentle suction holding pipette with its polar body aligned. The micro-injection needle penetrates the zona pellucida and oolemma, depositing the single sperm directly into the ooplasm.
Incubation & Blastocyst Transfer
Injected eggs are cultured in tri-gas incubators. Fertilization is verified 16–18 hours later by the presence of two pronuclei (2PN). Healthy blastocysts are then transferred into the prepared uterine cavity.
When is ICSI Specifically Recommended?
Who Should Consider This Treatment?
Essential for severe male infertility (extremely low sperm count, poor motility, high teratozoospermia), previous failed fertilization in conventional IVF, or surgically extracted testicular sperm.
Frequently Asked Questions
Physiological ICSI (PICSI) is an advanced method of selecting sperm for ICSI. It uses a hyaluronic acid gel to select mature, high-quality sperm that bind to the gel, mimicking natural selection.
ICSI is recommended for low sperm count, poor sperm motility, high abnormal morphology, previously failed fertilization, or when using surgically retrieved or frozen sperm.
Intracytoplasmic Sperm Injection (ICSI) is a technique where a single, healthy sperm is selected and injected directly into the center of the egg, bypassing the need for the sperm to swim.
